Put 1 T of vinegar in your measuring cup and add enough regular milk to equal 1 cup. This will make sour milk after standing for a few mintues. I have also heard you can use lemon juice instead of the vinegar.

What do you need it for? Sure, you can add acid by adding lemon juice or vinegar to milk, but you won't get the benefits of using real cultured buttermilk - which is cultures and flavor.
If you don't have buttermilk on hand I recommend you replace it with either sour cream or plain yogurt. Both have flavor and cultures and can be interchanged very easily.
